RESUMO

@#pdr5 and snq2 gene knockout was constructed by overlap PCR, and the effects of pdr5 and snq2 mutations on the accuracy and sensitivity of RNR2 promoter-regulated yeast cell sensors in detecting genotoxic compounds were studied. The yeast cell sensors of wild-type, single-gene mutation of pdr5, single-gene mutation of snq2, and double-gene mutation of pdr5 and snq2 were studied. The cell growth inhibition and the fluorescence induction factor of the yeast cell sensors exposed to a series of concentrations of methyl methanesulfonate(MMS), ethyl methanesulfonate(EMS), cisplatin, 4-nitroquinoline-N-oxide(4NOQ), 5-fluorouracil(5-FU), hydroxyurea, salicylic acid and glucose solution were investigated. The results showed that overlap PCR method could efficiently construct the mutant yeast cell sensor. The accuracy of cell sensors of single-gene mutation of snq2 and double-gene mutation of pdr5 and snq2 were both 100%, higher than that of cell sensors of wild-type and single-gene mutation of pdr5 (87.5%). The yeast cell sensor of double-gene mutation of pdr5 and snq2 showed the highest sensitivity in detecting genotoxicity. This study provides guidance for the construction of high accuracy and sensitivity yeast cell sensor, and foundation for further functional research of yeast cell membrane transporter gene pdr5 and snq2.

Objective@#To explore the association between drinking behavior and self injury behavior in adolescents.@*Methods@#A total of 9 247 students from 4 middle schools were investigated. Drinking behavior and self injury behavior were collected from questionnaire survey. Univariate and multivariate Logistic regression analysis were used to analyze the relationship between drinking behavior and self injury behavior.@*Results@#Among the 9 247 middle school students, 52.8% reported ever drinking, 24.9% reported drinking behavior in the past 30 days, and 14.6% reported been drunk in the past year. The average age of drinking for the first time was 12.47±3.05. About 47.2% of the participants had self injury behavior. Male with younger drinking age ( OR =1.52), had been drunken ( OR =1.35) and frequent drinking ( OR =1.54) increased the incidence of self injury. Female reported drinking at younger age ( OR =1.69), had been drunk ( OR =1.82) and lived in cities and towns ( OR =1.20) had a higher risk of self injury.@*Conclusion@#Drinking at younger age, heavy and frequent drinking are associated with higher risk of self injury in adolescents in sex specific fashion.

@#Objective: To investigate the role of momordica protein MAP30 in multiple myeloma (MM) and the possible mechanism. Methods: Human myeloma RPMI-8226, NCI-H929 and U266 cells were treated with MAP30 at different concentration (1-10 μmol/L) and then the proliferation rates of cells were detected by CCK-8 assay.Annexin V/PI flow cytometry was used to evaluate the apoptosis rate of myeloma cells, and the expressions of apoptosis-related protein (PARP), autophagy-related proteins (LC3II, P62) andAkt/mTOR pathway-related proteins in multiple myeloma cells were also detected via Wb. The changes in cell proliferation, apoptosis and autophagy after the treatment of MAP30 combined with autophagy agonist rapamycin (Rap) or autophagy inhibitor bafilomycin (Baf) were observed by CCK-8, flow cytometry and Wb, respectively. Results: MAP30 (1-10 μmol/L) inhibited the proliferation of myeloma cells in a time- and dose-dependent manner (P<0.05 or P<0.01). With MAP30 acting on myeloma cells alone, the apoptosis and autophagy of MM cells, as well as the expression of PARP cleavage and LC3II increased while the expression of P62 decreased significantly (all P< 0.05 or P<0.01). After being treated with MAP30+Baf, compared with MAP30 treatment alone, the cell proliferation was remarkably enhanced while cell apoptosis and cell autophagy were suppressed, besides, the expression of PARP cleavage and LC3II were decreased and P62 level was augmented (all P<0.05 or P<0.01). Conversely, after being treated with MAP30+Baf, compared with MAP30 treatment or Baf treatment alone, cell proliferation and P62 level were reduced, while apoptosis and autophagy as well as the expressions of PARP cleavage and LC3II level were increased (all P<0.05 or P<0.01). The expressions of p-AKT and p-mTOR were significantly reduced with the effect of MAP30 on myeloma cells (all P<0.05). Conclusion: MAP30 can promote the apoptosis and autophagy of myeloma cells throughAKT/mTOR pathway, which may provide a new therapeutic strategy for treatment of multiple myeloma.

Peripheral arterial disease (PAD) can impair healing of diabetic foot (DF) in patients with diabetes mellitus. To determine whether carotid intima-media thickness (CIMT) can predict lower limb arterial lesions in patients with DF, this cross-sectional study enrolled patients with DF at West China Hospital (China) between January 2012 and December 2015. Ultrasonography was used to measure CIMT, assess the internal carotid arteries (ICA) for plaques, and evaluate lower limb segmental arteries for stenosis. The optimal CIMT cutoff for detecting lower limb PAD was determined by receiver operating characteristic (ROC) curve analysis. Factors associated with PAD were identified by logistic regression analyses. A total of 167 patients (mean age: 69.7±10.3 years; 102 men) were included. Patients with PAD were older and had higher levels of total cholesterol and low-density lipoprotein than patients without PAD (P<0.05). The area under the ROC curve was 0.747 (P<0.001). At the optimal CIMT cutoff of 0.71 mm, the sensitivity, specificity, positive predictive value, and negative predictive value were 79.65, 61.11, 81.08, and 58.93%, respectively. Compared with those without PAD, more patients with PAD had CIMT ≥0.71 mm (79.65 vs 38.89%; P<0.001) and ICA plaques (66.37vs 11.11%; P<0.001). Multivariate logistic regression revealed that age (odds ratio [OR]: 1.118; 95% confidence interval [95%CI]: 1.056-1.183; P<0.001), ICA plaques (OR: 13.452; 95%CI: 4.450-40.662; P<0.001), and CIMT ≥0.71 mm (OR: 2.802; 95%CI: 1.092-7.188; P=0.032) were associated with PAD.CIMT may be a surrogate marker of PAD in patients with DF.

@#The esophageal disease is a major clinical disease. The esophageal stent has extensive clinical applications in the treatment of esophageal diseases. However, the clinical application of esophageal stent is limited, because there are lots of complications after implantation of esophageal stent. Biodegradable esophageal stent has two advantages: biodegradability and good histocompatibility. It is expected to solve a variety of complications of esophageal stent and provide a new choice for the treatment of esophageal diseases. Standardized esophageal stents are not fully applicable to all patients. The application of 3D printing technology in the manufacture of biodegradable esophageal stent can realize the individualized treatment of esophageal stent. And meanwhile, the 3D printing technology can reduce the manufacturing cost of the stent. This review aimed to summarize and discuss the application of esophageal stent, the current research status and prospect of biodegradable esophageal stent and the prospect of 3D printing technology in degradable esophageal stent, hoping to provide evidence and perspectives for the research of biodegradable esophageal stent.

Objective To observe the clinical efficacy of scalp acupuncture combined with electroacupuncture in the treatment of aphasia after ischemic stroke. Methods Totally 90 patients with aphasia after ischemic stroke were randomly divided into the control group and the treatment group, with 45 cases in each group. The control group received Schuell language training, 30 min each time, while the treatment group received scalp acupuncture plus electroacupuncture (Yifeng, Baihui, Houxi and Tongli), 20 min each time. The treatment for both group was 5 times a week and lasted for 16 weeks. The clinical efficacy was evaluated by aphasia degree, Chinese functional communication profile (CFCP) score and Western aphasia battery (WAB). Results There was statistical significance between the treatment group and the control group in the level of aphasia degree (P<0.05). Compared with before treatment, the total scores of CFCP and WAB in both groups increased, with statistical significance (P<0.05). The total scores of CFCP and WAB in treatment group were higher than control group, with statistical significance (P<0.05). The total effective rate of the therapeutic effect was 91.11% (41/45) in the treatment group and 75.56% (34/45) in the control group, with statistical significance (P<0.05). Conclusion Scalp acupuncture combined with electroacupuncture can obviously improve the language understanding and expression ability and language communication ability of aphasic patients after ischemic stroke.

<p><b>OBJECTIVE</b>To study effect of shoulder joint function after rotator cuff repair of polylactic acid absorbable membrane.</p><p><b>METHODS</b>From September 2015 to December 2016, 50 patients diagnosed with rotator cuff tear were selected and divided into treatment group and control group. There were 25 patients in control group, including 12 males and 13 females, with an average age of (48.7±3.5) years old, who received simple arthroscopic rotator cuff repair. There were 25 patients in treatment group, including 11 males and 14 females, with an average age of(49.2±4.1) years old, who performed arthroscopic rotator cuff repair with implanting polylactic acid absorbable membraneon shoulder of rotator cuff. Preoperative and postoperative VAS score, ASES score and UCLA score were recorded and compared between two groups.</p><p><b>RESULTS</b>At 6 months after operation, preoperative VAS score in control group was 5.48±1.12, and decreased as 1.28±0.84 after operation; ASES score before operation was 52.24±4.64, and improved to 86.92±3.20 after operation;preoperative UCLA score improved from 14.36±1.89 before operation to 30.72±1.28 after operation. In treatment group, VAS score decreased from 5.36±1.32 before operation to 1.40±0.71 after operation;preoperative ASES score was 51.04±4.09, and improved to 88.96±2.79 after operation; UCLA score improved from 15.12±1.81 before operation to 32.12±1.33 after operation. There was no significant difference in VAS score between two groups, and ASES score, UCLA score in treatment group was obviously better than control group.</p><p><b>CONCLUSIONS</b>Application of polylactic acid absorbable medical membrane could obviously improve shoulder function, and effectively prevent acromion adhesion after arthroscopic rotator cuff repair.</p>

Objective To establish an animal model of chronic nonbacterial prostatitis induced by chemical sub-stances, and provide a reliable animal model and evaluation method for pathological mechanism and pharmaceutical re-search of chronic nonbacterial prostatitis. Methods SD male rats were randomly divided into control group and model groups A, B and C. The three model groups were respectively treated with 20, 50 and 100 μL of 1% sterile carrageenan, injected into the left and right ventral lobes of rat prostate. The control group was injected 50μL sterile normal saline. The rats were sacrificed at 7 days after carrageenan injection, and the anatomical changes were analyzed, and the prostate in-dex, leukocyte count, the histology of prostate, and the protein expressions of TNF-α, NF-κB, IKKα, p-IKB-α and COX-2 were analyzed. Results Compared with the sham operation group, the softness of prostate tissue of groups A, B and C was decreased, the elasticity of the prostatic tissue was weakened, and the prostate tissues of model groups were ad-hered to surrounding tissues. The total number of leukocytes and the prostate index of model groups A, B and C were signif-icantly increased (P<0. 01), by 21. 1%, 61. 7% and 72. 7%, respectively. The total increase rate of white blood cell in the model groups A, B and C was 75%, 103. 6% and 114. 8%, respectively. Pathological examination showed that the in-terstitial edema of the prostate of model group A was minimal, but obvious in the groups B and C. Moreover, in the group C, the prostate atrophy was obvious, and some of the glands were degenerated and necrotic. The protein expression levels of NF-κB, IKKα, p-IKB-α, TNF-α, and COX-2 in the prostate tissue were increased to a different extent in all model groups. Conclusions Inflammatory reactions can be induced by injecting 20, 50 or 100 μL of 1% sterile carrag-eenan into the right and left ventral lobes of the rat prostate. However, the 20μL dose is too small, inducing only weak in-flammatory response, with considerable operation error, while the dose of 100μL induced excessive inflammatory response, even rat death. The dose of of 50 μL injection is most suitable to establish rat models of nonbacterial prostatitis, showing apparent activation of NF-κB inflammatory signaling pathway.

<p><b>OBJECTIVE</b>To evaluate the clinical effects of over 5 year follow up of unilateral pedicle screw fixation with transforaminal lumbar interbody fusion(TLIF) in treating lumbar degenerative diseases.</p><p><b>METHODS</b>The clinical data of 24 patients with lumbar degenerative disease underwent unilateral pedicle screw fixation with transforaminal lumbar interbody fusion from March 2007 to October 2009, were retrospectively analyzed. There were 13 males and 11 females, aged from 34 to 68 years old with an average of 52 years. Postoperative pain and functional results were analyzed by the visual analogue scale(VAS) and Oswestry Disability Index(ODI). Radiological examination was obtained for each patient to assess the height of intervertebral space, postoperative intervertebral fusion conditions and general complications.</p><p><b>RESULTS</b>All patients were followed up from 5 to 8 years with an average of 6.7 years. VAS scores of low back pain and leg pain decreased from preoperative 7.82±0.71, 8.42±1.24 to postoperative 1.87±0.81, 2.23±1.62, respectively(<0.05). ODI decreased from preoperative(53.42±8.26)% to postoperative(12.45±7.67)%(<0.05). Postoperative intervertebral space height in different segments were improved than preoperative(<0.05), but in final follow up it was decreased than 3 months after operation(<0.05). There was no significant difference in the postoperative intervertebral space height between the operated side and non operated side. But at final follow up, the intervertebral space height of non operated side was obviously loss. At final follow up, the fusion rate was 95.8%. The incidence of adjacent segment degeneration was 45.8%. The paraspinal muscle fibrosis incidence was 8.3%. No complications such as secondary scoliosis, intervertebral height loss, cage slippage, screw loosening and internal fixation breakage were found.</p><p><b>CONCLUSIONS</b>Unilateral pedicle screw fixation with TLIF is a satisfactory method and can obtain good effects in treating lumbar degenerative diseases according to over 5 year follow up, however, its indications should be well considered. But the problems such as intervertebral space height of operated side loss and adjacent segment degeneration after unilateral pedicle screw fixation need further clinical study.</p>

RESUMO

Post traumatic epilepsy (PTE) refers to the epileptic seizures after traumatic brain injury. Organic damage can be found by imaging examination, and abnormal electroencephalogram can be detected via electroencephalogram examination which has the similar location of the brain injury. PTE has the characteristics of low incidence, absence of case reports, and easy to exaggerate the state of illness, which add difficulties to the forensic identification. This paper reviews the status of epidemiology, pathogenesis, clinical treatment and forensic identification for PTE.

Our study aimed to investigate the measurement of frontal lobe volume and thalamic volume in fetuses with congenital heart disease [CHD] at different gestational weeks using three dimensional [3-D] ultrasonography and its clinical value. Then, 238 pregnant women who received obstetric ultrasonography in ultrasound department of Internal Medicine of our hospital were enrolled between March 2013 to April 2014. In this study, 85 fetuses were diagnosed to develop CHD by prenatal fetal echocardiography, and the other 153 fetuses were normal. Frontal lobe volume, thalamic volume and cerebral blood flow was determined by color Doppler ultrasonic diagnostic apparatus [type: GE Voluson E8]. The level of MCA-PI and CPR in CHD fetus group performed significantly lower than that in normal fetus group [P<0.05], but the level of UA-PI performed significantly higher than that in normal fetus group [P<0.05]. When gestational age <30 weeks, there was no significant difference in thalamic volume and frontal lobe volume between the two groups [P>0.05]; when gestational age >/= 30 weeks, the level of CHD fetus group performed significantly lower thalamic volume and frontal lobe volume than that in normal fetus group [P<0.05]. When gestational age <30 weeks, there was no significant difference in BPD, HC, and GA between the two groups [P>0.05]; when gestational age >/= 30 weeks, the level of BPD, HC and GA in CHD fetus group performed significantly lower than that in normal fetus group [P<0.05]. If gestational age <30 weeks, CHD performed a small impact on fetal frontal lobe volume and thalamic volume; if gestational age >/= 30 weeks, the level of frontal lobe volume and thalamic volume in fetuses with CHD performed significantly lower than that in normal fetuses

To facilitate the biodegradation of diesel oil, an oil biodegradation bacterial consortium was constructed. The alkane hydroxylase (alkB) gene of Pseudomonas putida GPo1 was constructed in a pCom8 expression vector, and the pCom8-GPo1 alkB plasmid was transformed into Escherichia coli DH5α. The AlkB protein was expressed by diesel oil induction and detected through SDS-polyacrylamide gel electrophoresis. The culture of the recombinant (pCom8-GPo1 alkB/E. coli DH5α) with the oil biodegradation bacterial consortium increased the degradation ratio of diesel oil at 24 h from 31% to 50%, and the facilitation rates were increased as the proportion of pCom8-GPo1 alkB/E. coli DH5α to the consortium increased. The results suggested that the expression of the GPo1 gene in E. coli DH5α could enhance the function of diesel oil degradation by the bacterial consortium.

<p><b>OBJECTIVE</b>To study the clinical value of cranial magnetic resonance imaging (MRI) in the diagnosis and treatment of central nervous system candidiasis (CNSC), which has no specific clinical manifestations and has no rapid and specific diagnostic tools.</p><p><b>METHODS</b>A retrospective analysis was performed on the clinical data of 10 children who were diagnosed with CNSC in Beijing Children's Hospital Affiliated to Capital Medical University between 2009 and 2013.</p><p><b>RESULTS</b>Nine of the 10 children underwent cranial MRI within 8 days after admission, and 5 of the 9 children underwent contrast-enhanced MRI at the same time. Eight of the 9 children showed the features of meningoencephalitis, and 6 cases were accompanied by varying degrees of brain atrophy; one case showed hydrocephalus and cerebral abscess, and another case showed leukoencephalopathy. Six cases were found to have the features of cerebral vasculitis after infection in the first MRI after admission, including cerebral infarction (2 cases), venous sinus thrombosis (3 cases), and Moyamoya disease (1 case). Infectious granulomatous lesions were confirmed by contrast-enhanced MRI in 3 cases. Given the clinical manifestations, 8 of the 9 cases were diagnosed as suspected CNSC after MRI, and 7 of these cases received antifungal therapy before the pathogen test results were returned. The lesions on MRI were improved in 6 cases after 3-4 weeks of antifungal treatment. All the 10 children were diagnosed with CNSC by positive cerebrospinal fluid culture results.</p><p><b>CONCLUSIONS</b>Cranial MRI, especially contrast-enhanced MRI, is of great significance for the diagnosis and treatment of CNSC. To confirm the guidance of MRI in the diagnosis and treatment of CNSC, further case-control studies are needed.</p>

<p><b>OBJECTIVE</b>To investigate the mRNA expression and promoter methylation status of p73 gene in the peripheral blood of children with Wilms' tumor (WT), and their relationship.</p><p><b>METHODS</b>Forty-five children with WT were selected as the case group, and 15 sex- and age- matched children (without malignancies) who visited the hospital for physical examination or other reasons were selected as the control group. Peripheral blood was collected from both groups. Real-time quantitative PCR and methylation-specific PCR were used to determine the mRNA expression level and promoter methylation status of p73 gene. Their relationship with clinicopathological features and the effect of promoter methylation on mRNA expression of p73 gene were analyzed in the case group.</p><p><b>RESULTS</b>The relative quantity (RQ) of p73 mRNA in the case group was significantly higher than in the control group (3.2 ± 0.9 vs 1.6 ± 1.1; P<0.01). The positive rate of p73 gene promoter methylation in the case group was significantly lower than in the control group (20% vs 73%; P<0.01). In the case group, the RQ of p73 mRNA was significantly higher in children with methylated p73 gene promoter than in those with unmethylated p73 gene promoter (P<0.01). In children with methylated p73 gene promoter, the RQ of p73 mRNA was significantly higher in the case group than in the control group (P<0.01). In children with unmethylated p73 gene promoter, there was no significant difference in RQ of p73 mRNA between the case and control groups (P=0.810).</p><p><b>CONCLUSIONS</b>Aberrant promoter methylation of p73 gene in peripheral blood is one of the gene expression regulations in children with WT, and it is related to the onset and development of WT. The p73 gene may play a role as oncogene in WT patients with p73 gene promoter methylation and mRNA overexpression is associated with promoter methylation status of p73 gene.</p>

RESUMO

<p><b>OBJECTIVE</b>To explore the safety testing evaluation index of breast-feeding by hepatitis B-positive mothers.</p><p><b>METHODS</b>HBV DNA from serum and breast milk of 252 hepatitis B-positive mothers were detected with the real-time quantitative PCR.</p><p><b>RESULTS</b>The total positive rate of HBV DNA in serum had no difference with that in breast milk in hepatitis B-positive mothers (P > 0.05). The positive rate of HBV DNA in serum and breast milk of positive HBeAg were significantly higher than that of hepatitis B-positive mothers with negative HBeAg (P < 0.01).</p><p><b>CONCLUSION</b>To detecte HBeAg and HBV DNA in serum and breast milk have important significance for guiding of breast feeding of hepatitis B-positive mothers.</p>

<p><b>BACKGROUND</b>Hepatoblastoma (HB) is a rare childhood tumor. We investigated the effect of intraoperative management of the intrahepatic major vessels in children with HB.</p><p><b>METHODS</b>Between April 2005 and August 2012, surgical resection was performed on 50 children with hepatoblastoma. These children were divided into a vessel-ligation group (n = 20) and a vessel-repair group (n = 30). In the vessel-ligation group, the intrahepatic major vessels were ligated and removed together with the tumor and the affected liver lobe/liver parenchyma. In the vessel-repair group, the affected intrahepatic major vessels were dissected and preserved as much as possible and the normal liver lobe/liver parenchyma and blood supply from these vessels were also preserved. The outcomes were analyzed by postoperative follow-up.</p><p><b>RESULTS</b>In the vessel-ligation group, two patients gave up surgery, six patients underwent palliative resection, and 12 patients underwent en bloc resection; four patients died of liver failure and eight patients fully recovered and were discharged. In the vessel-repair group, all 30 patients underwent en bloc resection and were discharged after satisfactory healing. After a follow-up time of 5 - 36 months (median: 20 months), two patient in the vessel-ligation group survived and 22 patients in the vessel-repair group survived.</p><p><b>CONCLUSIONS</b>Patients with HB can be successfully treated by tumor resection with vascular repair. This method prevents postoperative liver failure, ensures patient safety during the perioperative period, and allows for early chemotherapy.</p>

RESUMO

A pair of specific primers were designed according to the published 118 bp conserved sequence of polyhedrin gene of Bombyx mori cytoplasmic polyhedrosis virus (BmCPV) in GenBank and a serial dilutions of a recombinant plasmid were prepared and used to generate standard curves, to establish a real-time fluorescent quantitative PCR method for detection of BmCPV. The results showed that the linear relationship between virus concentration (X) and Ct (Y) was Y = -3. 582 lgX + 38.748 with the correlation coefficient R2 = 0999. The method was very sensitive, specific and reproducible. It can be applied in the rapid detection of BmCPV and the prevalence investigation of this disease.

Objective To summarize clinical,gene mutation and their families of 4 Chinese children with X-linked lymphoproliferative (XLP) disease.Methods The clinical records and 6 genes of immunodeficiency associated with Epstein-Barr(EBV) infection were summarized and the literatures were reviewed.Results The four cases were all boys younger than 3 years old,who had onset with fever.They were all treated with ganciclovir,plasma,intravenous immunoglobulin and methylprednisolone after hospitalization,but 3 cases had the features of fulminant or fatal infectious mononucleosis (FIM),whose progression of disease was getting worse and died of second hemophagocytic lymphohistiocytosis (HLH) in the end.The survival time after onset was about 20 days.One boy had the complications of HLH associated with EBV infection and drug-induced hypersensitivity syndrome,who was improved and discharged from hospital.Two cases had adverse family history in which brothers or cousins died at younger ages.EBV-CAIgM and EBV-DNA of the 4 cases were all positive,with the copy of EBV DNA > 108 copies/L.The results of the 6 genes of immunodeficiency associated with EBV infection of the 4 boys showed SH2D1A mutation.Mothers of 3 cases separately had the same SH2D1A mutation as her boy,while 1 mother refused to have the genes detected.Conclusions Patients who had the XLP were all male.Infants and young children under 5 years old usually had the features of FIM,with the complication of EBV associated HLH.Patients with XLP showed SH2D1A mutation.For male patients with FIM,especially those under 5 years,it is important to perform genetic detection early and to receive therapy as early as possible.

The study is to investigate the pharmacokinetics of S-1 capsule (tegafur, gimeracil and potassium oxonate capsule) in patients with advanced gastric cancer after single and multiple oral administration. Twelve patients with advanced gastric cancer were recruited to the study. The dose of S-1 for each patient was determined according to his/her body surface area (BSA). The dose for single administration was 60 mg every subject. The dose for multiple administration for one subject was as follows: 100 mg x d(-1) or 120 mg x d(-1), 28-days consecutive oral administration. The pharmacokinetic parameters of tegafur, 5-fluorouracil, gimeracil, potassium oxonate and uracil after single oral administration were as follows: (2,207 +/- 545), (220.0 +/- 68.2), (374.9 +/- 103.0), (110.5 +/- 100.8) and (831.1 +/- 199.9) ng x mL(-1) for Cmax; (11.8 +/- 3.8), (4.4 +/- 3.3), (7.8 +/- 5.1), (3.1 +/- 0.9) and (8.8 +/- 4.1) h for t1/2, respectively. After six days oral administration, the average steady state plasma concentrations (Cav) of tegafur, 5-fluorouracil, gimeracil, potassium oxonate and uracil were (2,425 +/- 1,172), (73.88 +/- 18.88), (162.6 +/- 70.8), (36.89 +/- 29.35) and (435.3 +/- 141.0) ng x mL(-1), respectively, and the degree of fluctuation (DF) were (1.0 +/- 0.2), (2.5 +/- 0.4), (3.1 +/- 0.8), (2.4 +/- 0.8) and (1.5 +/- 0.3), respectively. The cumulative urine excretion percentage of tegafur, 5-fluorouracil, gimeracil and potassium oxonate in urine within 48 h were (4.2 +/- 2.8) %, (4.7 +/- 1.6) %, (18.5 +/- 6.0) % and (1.7 +/- 1.2) %, repectively, after single oral administration of S-1. The results exhibited that tegafur had some drug accumulation observed, and gimeracil, potassium oxonate, 5-fluorouracil and uracil had no drug accumulation observed.
